生まれは明治5年3月25日（1872年5月2日）、東京府第二大区一小区内山下町一丁目一番屋敷で樋口則義と多喜の次女として生まれた。父が官吏だった影響で、早くから書や読み方を覚えている。幼少の頃は手毬や羽根つきなどの遊びではなく、草双紙などを読んでいた。一葉自身が日記に記したように、「曲亭馬琴『南総里見八犬伝』を3日で読破」したこともあった。
しかし、次兄が勘当されて一葉は同じ東京府内の下谷区御徒町へ移った。11月に上野元黒門町の私立青海学校に転校する。ここで初めて和歌を習う。しかし、母が女性に学業は不要だと考えていたため、高等科第四級を首席で卒業しても上級には進まず退学した。
