三田紀房は1958年1月4日に生まれた。岩手県北上市出身で、明治大学政治経済学部を卒業した。彼は剣道競技に興味があり、それが初期の作品「空を斬る」に影響を与えたと言われている。また、彼の故郷である岩手県が舞台になっている村上もとかの代表作である「六三四の剣」にも影響を与えている。
大学卒業後、西武百貨店に入社したが、1年後に退職して実父の家業に戻った。そこで2年後に父は病死し、その後多額の借金が残って資金繰りに苦しみ、漫画の制作・新人賞応募を決意した。
30歳の時、「さよならの贈り物」が講談社の第17回ちばてつや賞一般部門に入選し、その後は店番と漫画を描く二足のわらじを続けた。アシスタント経験がない彼は、学生時代にも創作活動をしていなかった。
スポーツ漫画や野球漫画で徐々に頭角を現したが、仕事は細々であったため生活はギリギリの状態であった。そんな中、「漫画ゴラク」の編集者の激励が転機となり、自らアンケート結果などを研究し「売れる漫画」のパターンを研究するようになった。
その後2003年より『モーニング』で連載を開始した「東大受験」をモチーフにした異色作『ドラゴン桜』が大ブレイクのきっかけとなり、2005年に第29回講談社漫画賞と平成17年度第9回文化庁メディア芸術祭マンガ部門優秀賞を受賞し、「東大受験」をテーマにした異色作『ドラゴン桜』がテレビドラマ化された。同作品の続編として2007年から2010年に『モーニング』で連載した『エンゼルバンク-ドラゴン桜外伝-』もテレビドラマ化されている。
その他にも「マネーの拳」、「銀のアンカー」を連載し、野球漫画「砂の栄冠」を連載。2015年から現在は戦争漫画「アルキメデスの大戦」を連載している。また2021年に連載終了した「ドラゴン桜2」も受験漫画である。
2026年4月には「ドラゴン桜財団」を設立し、全国の中高生らを対象に学年に応じて年間30万～60万円を支給する奨学金制度や学習支援プログラムを提供している。

After that, ``Dragon Zakura'', a unique work with the theme of ``Tokyo University entrance exams'', which began serialization in ``Morning'' in 2003, became a big breakout, winning the 29th Kodansha Manga Award in 2005 and the 9th Japan Media Arts Festival Excellence Award in the Manga Division in 2005, and his unique work with ``Tokyo University entrance exams'' as a theme was made into a TV drama. A sequel to the same work, ``Angel Bank - Dragon Sakura Gaiden'', which was serialized in ``Morning'' from 2007 to 2010, has also been made into a TV drama.
